They never exactly said why Isabelle aged. However Lily aged due to Isabelle's aging. Isabelle was basically sucking the life out of Lily, which was written in because the actress who played Lily was not returnig.

They aged Isabelle because it was part of the plot that SHE be one of the main protagonists of the future of the 4400's, as an adult. They wanted to go ahead and skip ahead to the part of the story WHERE as an adult she's a main character. The could of course have dragged that out for years and years, aging her a couple years at a time with each season. There wasn't much way to make her grow up over night without just going ahead and making her grow up over night. It wasn't necessary to have also made Lily grow old, they didn't age and kill Richard. There was apparently contract issues with the actress that plays Lily and she was leaving the show and IS already on some other show. They chose to kill her off rather than supply some other explanation for her leaving.

I can empathize with that choice. I employ a few people. I never hire back people who have quit me unless they were REALLY REALLY REALLY nice about it. Maybe Laura wasn't nice about it. Just saying that because some people felt that rather than killing her off the producers could have sent her off to an island or something leaving her the option to come back. I agree with the producers, quit me once, shame on you, quit me twice, shame on me.
